It's very easy. My first build took around an hour to assemble. Now, I can do it in half that. Probably less, even.
Basically, everything only goes together one way. Everything has different types of connectors that you can only plug into one place. There aren't really as many wiring connections as most people think. Figuring out the PSU cables the first time is probably the most difficult part. But it's still quite simple, especially now that many PSUs come with labeled connectors.
1. Get your stuff out of the boxes and ready to install
2. Open up the case and install standoffs if you haven't already
3. Install CPU and CPU cooler onto motherboard
4. Install RAM onto mobo
5. Put power supply in case and screw it in
6. Put optical drives and hard drives in case
7. Put mobo I/O shield into case opening
8. Put mobo into case
9. Connect your case wiring (wires for on/reset button, power/hard drive LEDs, USB, audio, etc)
10. Install your graphics card and other PCI/PCIe cards
11. Run your power cables, connect SATA cables
That's pretty much all there is to it. Then you just install your choice of OS, install drivers, and you're good to go
Edit: Doesn't necessarily have to be in exactly the order I listed them, but that's the order I like to do it in.
